TNFRSF14
El mediador de entrada del herpesvirus (HVEM), también conocido como miembro 14 de la superfamilia del receptor del factor de necrosis tumoral (TNFRSF14), es un receptor de superficie celular humana de la superfamilia de receptores de TNF [1][2][3][4] codificado por el gen TNFRSF14.[3]
Nomenclatura
[editar]Esta proteína se conocía originalmente como mediador A de entrada del herpesvirus ( HveA ); HveB y HveC son proteínas estructuralmente no relacionadas de la superfamilia de inmunoglobulinas. También se le conoce como CD270 en el grupo de clasificación de diferenciación. Además, también se le conoce como ATAR (otro receptor asociado a TRAF).
Función
[editar]La proteína codificada por este gen es miembro de la superfamilia de receptores de TNF. Se descubrió que la región citoplasmática de este receptor se une a varios miembros de la familia del factor asociado al receptor de TNF (TRAF), que pueden mediar en las vías de transducción de señales que activan la respuesta inmune.[3]
En las células melanocíticas, la expresión del gen TNFRSF14 puede estar regulada por MITF.[5]
Interacciones
[editar]Se ha demostrado que TNFRSF14 interactúa con TRAF2,[6][7] TNFSF14 [8][9] y TRAF5.[6] [7]
Relevancia clínica
[editar]Las mutaciones en este gen se han asociado de forma recurrente con casos de linfoma difuso de células B grandes [10][11] y linfoma folicular de tipo pediátrico.[12]
Este receptor fue identificado como un mediador celular de la entrada del virus del herpes simple (VHS). Se ha demostrado que la unión de la glicoproteína D (gD) de la envoltura viral del VHS a esta proteína receptora es parte del mecanismo de entrada viral.[3]
