The Janus kinase (JAK)–signal transducer and activator of transcription (STAT) is an important immune system pathway that research over the past 20 years has shown is also involved in skin barrier function and disorders. As such, various treatments that influence this pathway have been approved by the US Food and Drug Administration (FDA) to treat atopic dermatitis, a chronic skin disease with extensive quality-of-life effects, whose prevalence is increasing throughout the population. Understanding how the JAK-STAT pathway influences the pathogenesis of atopic dermatitis and how therapies that modulate it affect both the disease and those who suffer from it is key to providing patients a variety of options for treatment.
